GDC-0449, Vismodegib is a potent inhibitor of Smoothened (SMO), a key mediator of Hedgehog (Hh) signaling pathway. GDC-0449, Vismodegib inhibition of SMO renders the transcription factor GLI inactive, thus preventing the expression of genes that mediate the role of Hh on tumor growth.
