"作为后基因组时代表达克隆和蛋白质纯化技术*重要的供应商之一，德国IBA公司开发了一个称为Strep-tag的通用技术平台。这种快速、合算、多样性地生产和使用重组蛋白的技术平台是IBA与TU Munich的Arne Skerra博士教授密切合作开发而成。除了围绕Strep-tag的大型产品组合外，IBA还**提供应用该技术的蛋白质生产定制服务。Strep-tag技术开发的原理是众所周知的生物素（biotin）和抗生物素蛋白链菌素（streptavidin）之间的结合反应。为了将这种牢固的相互作用用于蛋白质纯化用途，研究者们发现需要一个肽，该肽与重组蛋白质融合后，能够结合在streptavidin的生物素结合口袋，从而作为纯化tag。*后研究者们成功设计出一个只由8个氨基酸(Trp-Ser-His-Pro-Gln-Phe-Glu-Lys)构成的短序列并称之为Strep-tag II。为了优化结合属性，streptavidin也被设计成为Strep-Tactin。如此，一对优化的结合伙伴被发现了：Strep-tag / Strep-Tactin系统现已成为*被广泛应用的亲和层析系统之一。Strep-Tactin与多种不同的基质偶联，使Strep-tag融合蛋白得以在生理条件下亲和纯化。与其他tag相比，这些温和的纯化参数能保存蛋白质的生物活性，并仅经一步层析后即可产出超过99%的纯度。
