Immortalization Method Transfection preformed with pSV3-SV40-T antigen plasmid DNA
Description Immortalized Mouse CD81−/− Macrophage Cells (ASD1) (Cat. No. T0138) is established from the long bones of CD81 -/- mice, which were immortalized with the SV40 large T antigen and underwent limiting dilution cloning. Immortalized Mouse CD81+/− Macrophage Cells (2BSD1.10) was generated from transfecting the ASD1 cells with the pCDNA3.1/Hygro vector carrying the CD81 gene. The cell line offers users the ability to conduct experiments with a reliable source of immortal cells in vitro. The CD81 gene has associations with malignancies as it is localized within the tumor-suppressor gene region. Hence valuable for studies in regards to cancer.
Procedure Overview

Propagation Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow III medium available in abm, Cat. No. TM003. To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999) to final concentration of 6%, 6% Nu-Serum (Corning™ 355100), and 1% Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
