Description SSAO (semicarbazide-sensitive amine oxidase) has been associated with diabetes, Alzheimer’s, atherosclerosis, and stroke. Thus, it is necessary to have a reliable source of cells which expresses the desired protein for studies in its role and activity in pathways. The hSSAO/VAP-1 A7r5 Stable Cell Line stably expresses SSAO/VAP-1 in the plasma membrane, and releases SSAO into the culture medium. This cell line does not exhibit MAO-A nor MAO-B activities. It is a valuable source for studies in SSAO/VAP-1 membrane-bound form enzyme as a therapeutic target.
Procedure Overview
Recommended Seeding Density Thaw entire contents into an appropriate T25 flask as specified in the Propagation instructions.
Applications For Research Use Only
Over Expression Human SSAO/VAP-1
Propagation Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow III medium available from ABM (TM003). To make the completed growth medium, add the following components to the base medium to a final concentration of 10% fetal bovine serum (TM999), 2 mM L-glutamine (G275) and Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.

Quality Control 1) Selection via antibiotic G418; 2) Western blot used to determine presence of SSAO; 3) Activity of SSAO, MAO-A, and MAO-B was detected by radiochemical methods
